Agar Plug Inocula Affect Accuracy of Cultural Tests of Inhibition of Fungi By Chemicals

Past studies show growth of Hypoxylon pruinatum in plates of Difco malt agar was more closely related to the type of inoculum than the inhibiting potency of the test medium. The present study was on development of fungi in plates of Difco malt agar containing different chemical inhibitors. Concentrations of three chemicals that resulted in total inhibition of the fungi from the agar plug inocula in contact with the test media are reported. In all instances, however, fungi did develop on the test media from the inocula on the sterile cover slips. Further mycelial growth continued after the cover slops and inocula were removed. It therefore appears to be undesirable to use agar plug inoculum in contact with the test medium in investigations to determine the concentration of chemicals for inhibition of fungus growth.
